example being I have 2 usb 3.0 ports on my setup only 2 x16 for gpu's
I guess technically it should work but would the speed be there?

The photo shows a PCIe 1x USB extender and has absolutely nothing to do with the 'normal' USB ports of your computer.

The only thing you need is a free PCIe port.
